Non-mutational mechanism of Platinum resistance in ovarian cancer

Ovarian cancer is the eight most common malignancies in women worldwide. First line therapy
combines surgery with platinum-based chemotherapy. Despite the initial response, progressive
develop of drug resistance becomes more frequent and difficult to avoid. Overcoming Pt-
resistance is one of the main unmet clinical needs for EOC patient. In particular we will investigate
the role of USP1, a deubiquitinase protein that regulates processes in response to DNA damage
repair and Epithelial to Mesenchymal transition, already identified as one of the proteins possibly
involved in PT-resistance. Thus, the aim of my PhD project is to investigate the role of USP1 in
Platinum response and resistance, identifying new possible therapies to overcome EOC patients
recurrences. For this instance, we will explore the transcriptional, translational and post-
translational regulation of USP1, comparing its normal activity with our Pt res cell models, trying to
identify new possible mechanism involved in platinum resistance.
