Role of ROS/RNS induced by cancer drugs in the resistance and recurrence of pancreatic cancer

The aim of this project is to study the role of reactive oxygen and nitrogen species and nitric oxide
(ROS /RNS, NO) generated by cancer treatments in the development of drug resistance in pancreatic ductal adenocarcinoma (PDAC). Pancreatic adenocarcinoma is a serious disease with a poor prognosis and a survival rate of about 10%, poorly responsive to conventional therapies due to its advanced stage. Surgical resection followed by chemotherapy with paclitaxel or gemcitabine is the first choice. The objectives of this study are, first, to develop and characterize a gemcitabine- resistant cell line that will undergo repeated low-dose treatments and, second, to evaluate the role of ROS/RNS and NO in this process. In addition, it is of interest to investigate the possibility of modulating the concentration of reactive species (using antioxidants or NO synthase (iNOS) inhibitors or NO donors) to re-sensitize the resistant line to cancer treatment.
