Molecular approach for mRNA profiling of antisense ONs in Huntington disease and cancer cellular models

The PhD project will mostly focus on the characterization of the biological effects of ONs using different Huntington’s Disease and Cancer cellular models, with a main interest on mitochondria. The concern in this organelle comes from the association of mitochondrial DNA damage with mitochondrial dysfunction, which can lead to the development of a variety of human diseases. Currently, there are no proper mitochondrial therapies for such pathologies and so, successful therapies and the development of efficient mitochondrial drug delivery systems are needed.
In particular, it is plan to evaluate how antisense oligonucleotide therapy affect cell toxicity, gene and protein expression, by studying cell viability, performing gene array analysis and doing pathway evaluation, among others.
As part of the project a secondment in CNRS in Marseille, FR (The CNRS | CNRS) is planned for carrying out the validation of the pathways activated in response to ONs treatment as well as an extension in the knowledge on gene ontology analysis.
